Objective: To investigate the proliferation influence of tumor from gastric cancer orthotopic transplanted nude mouse and discuss the mechanism.

Methods: Gastric cancer orthotopic transplanted nude mouse mode with human gastric cancer cell line SGC-7901 were prepared and transfected in vivo. The tumor tissues were taken for pathological examination after complete transfection. MTT colorimetry was used to detect cell proliferation of gastric cancer orthotopic transplanted nude mouse after primary culture. The cell cycle was detected by flow cytometry. The expression of ZNF-139, PCNA and Cyclin D mRNA were detected by RT-PCR.

Results: The growth of human gastric cancer orthotopic transplanted tumor in nude slowed down significantly after transfection. The MTT results showed that cell growth inhibition rate reached the highest level at 48 hours, the values were 31.5%. After gastric cancer orthotopic transplanted tumor transfection, Cells percentages in G0/G1 phase were more than the untransfected group, whereas the cells percentages in S phase were less than the untransfected group. The results of RT-PCR showed that the expression of ZNF-139, PCNA and Cyclin D mRNA in tumor of gastric cancer orthotopic transplanted nude mouse were inhibited significantly.

Conclusions: The proliferation of tumor in gastric cancer orthotopic transplanted nude mouse slowed down after transfection by SiRNA. Its mechanism which lies in ZNF-139 is gene regulated factor in transfection level, which may decline genetic expression of PCNA and CyclinD1 and further influence the progress of cell cycle, slow down the tumor cell proliferation and play role in anti-tumor after silence.
