AB23. The effects of xinjialiangfu formula on Bax and Bcl-2 expression of transplanted human gastric cancer cell line

Objective: To observe the effect of tumor growth of transplanted human gastric cancer (SGC-7901) on nude mice and expressions of Bcl-2 and Bax proteins in tumor tissues.
Methods: Animal models with human gastric cancer xenografts were established and randomly divided into 6 groups; namely model control group, 5-Fu group, Modified Prescription Liangfu high-dose group, Modified Prescription Liangfu medium-dose group, Modified Prescription Liangfu low-dose group and Combined drug group (Modified Prescription Liangfu medium-dose group and 5-FU group). Modified Prescription Liangfu groups of high, medium and low dosage were administered Modified Prescription Liangfu at the amount of 10 g/kg, 5 g/kg and 2.5 g/kg respectively; while 5-Fu group was administered 5-Fu at a dose of 17 mg/kg, and the model control group was given equivalent sterile normal saline. Combined drug group was administered Modified Prescription Liangfu medium-dose group and 5-Fu group. All mice were sacrificed after 10 days of continuous drug and water delivery. The tumor blocks were separated, weighed, embedded in paraffin, dissected into slices, and Bax and Bcl-2 expression was detected by immunohistochemical method.
Results: The tumor inhibition rate of Combined drug group was 70.07%. The comparison between the model group and Combined drug group in tumor weight was significant statistically (P<0.05); Compared with 5-FU group and Liangfu groups of high, Combined drug group tumor inhibition rate was increased significantly (P<0.05). Compared to the model group, Combined drug group could increase the expression of Bax protein and decrease the expression of Bcl-2 protein (P<0.05, P<0.01); Compared with 5-FU and t Liangfu groups of high, Combined drug group increase the expression of Bax protein significantly (P<0.05), decrease the expression of Bcl-2 protein significantly (P<0.05).
Conclusion: The tumor inhibition rate of Combined drug group is higher than Modified Prescription Liangfu high-dose group and 5-FU group, and decrease expression of Bcl-2, Bax expression in terms of increase is better than Modified Prescription Liangfu high-dose group and 5-FU group, shows the two drugs have a certain inhibitory effect.
