Objective:
To investigate the expression of zinc finger protein 139 (ZNF139) and its relationship with matrix metalloproteinase-2 (MMP-2), MMP-7 gene in the primary tumor and metastases of orthopotic implantation nude mice.
Methods:
Orthopotic implantation nude mice with human gastric carcinoma cells was established. Then reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemical method (SP) were used to detect the expression of ZNF139, MMP-2, MMP-7 in orthotopic transplantation tumor, liver metastases, spleen metastasis, peritoneal metastasis, lymph node metastasis.
Results:
All models got tumors successfully (93.33%, 28/30). Hepatic metastases (HM) were found in 75.00% (21/28) models, splenic metastases (SM) in 67.86% (19/28) models, peritoneal metastases (PM) in 85.71% (24/28) models, lymph node metastases (LNM) in 39.29% (11/28) models. Conclusions:
Models of orthopotic implantation nude mice could stimulate the transfer process. Invasion and metastasis was stronger in metastases than in PT, and ZNF139 may be involved in this process by regulating MMP-2, MMP-7.
